At room temperature, procaine hydrochloride is a fine white needle-like crystal
or crystalline powder, odorless, slightly bitter and tingling, easily soluble
in water, soluble in ethanol, slightly soluble in chloroform, almost insoluble
in ether.In the structure of procaine hydrochloride, the ester bond can be
hydrolyzed to form p-aminobenzoic acid and diethylaminoethanol. Under certain
conditions, p-aminobenzoic acid can be further decarboxylated to form toxic
aniline.The structure of procaine hydrochloride has aromatic amino group, which
is easy to oxidize and change color. The oxidation and change color can be
accelerated by pH value, temperature, ultraviolet light, oxygen and metal ions.
Hydrochloric acid procaine is a local anesthetic, can temporarily block the
conduction of nerve fibers and has an anesthetic effect, strong effect, low
toxicity, and no addiction, but on the skin, mucous membrane penetration is
weak, not suitable for surface anesthesia, clinical mainly used for
infiltration, spine and conduction anesthesia.
The aqueous solution of this product is stable in pH3.0 ~ 5.0.Therefore, when
preparing procaine hydrochloride injection, it is generally necessary to add
acid to adjust pH3.3 ~ 5.5, and strictly control the sterilization temperature
and time. It is advisable to circulate steam for sterilization at 100℃ for
30min, and attention should be paid to shading and airtight preservation.This
product structure of the aromatic amino in dilute hydrochloric acid, with
sodium nitrite to generate diazo salt, coupled with basic β-naphthol reaction,
the generation of scarlet azo compounds.
